Priyamaina Neeku is a romantic drama directed by Balasekharan, starring Tarun as Ganesh and Sneha as Sandhya. The story begins with Ganesh, a cheerful and helpful college student whose life is centred around his friends. One day, he stumbles upon a red diary in his father's auction shop. This diary belongs to Sandhya, his former neighbour. The first half of the film is narrated through the entries in Sandhya's diary. Sandhya, a shy and reserved girl, lives opposite Ganesh's sister, Aishwarya. She secretly falls in love with Ganesh after being captivated by his charm and skillful guitar playing. Ganesh remains unaware of her feelings as she never expresses them directly. Her younger sister, Sirisha (Preetha Vijayakumar), comes home for holidays and, upon learning of Sandhya's affection for Ganesh, grows jealous. Sirisha cunningly pretends to help Sandhya but instead tries to win Ganesh over for herself. This leads to a moment where Sirisha proposes to Ganesh, but he rejects her. Sandhya witnesses this and feels betrayed by her sister, yet she still hesitates to express her love to Ganesh due to fear of rejection. Later, Sandhya's family moves to Visakhapatnam, and in the process, her diary gets misplaced. After Ganesh discovers the diary and realizes Sandhya is his secret admirer, he acknowledges the purity of her love. He sets out to find her in Visakhapatnam, accompanied by his friend Chacha (Venu Madhav). He ends up befriending Raghu (Sivaji) and staying at his home. In a twist of fate, Raghu becomes engaged to an unwilling Sandhya. Ganesh, who doesn't know how Sandhya looks, realizes that Raghu is marrying her. Ganesh is torn between his friendship and his love. The rest of the film explores how Sirisha tries to rectify her previous mistakes and unites Sandhya and Ganesh.
